STUDY ON MONOCLONAL ANTIBODY AGAINST DIFFERENTHUMAN CYTOMEGALOVIRUS POLYPEPTIDES

  • Aim:Nonionic detergent solubilised Polypepdides from human cytomegalovirus virions. Were used as im-munogens to preduce monoclonal antibodies (McAb). Method: 13 hybrid cells which secrete hybrdization technique McAbs a-gainst different human cytomegalovirus (HCMV) polypeptides were established by cell fusion. 6 hybrid cells (7B4. 7D7.7E7. 7E11.8B8. 8D6) were selected for further. Results: All subclass of the 6 McAb are IgG1 type (molecular weight of heavy chain and light chain is separately 50KD. 25KD). Indrect immunofluorescence reation of 6 McAb with HCMV infected cells at different time showed that McAb 8B8 is against the immediate early antigen of HCMV, while McAb 7B4' 7D7. 7E7.7E11. 8D6 are against the late antigen of HCMV. Immunoblot analysis revealed 6 McAb are directed against different human cytomegalovirus polypeptides with molecular weight of approximately 46. 150. 38. 51.72.65 KD. Conclusion: 6 McAb may by used to diagnose HCMV.
