重组人骨形态发生蛋白10的表达纯化及其心脏保护功能

Expression and purification of recombinant human bone morphogenetic protein 10 and it cardiac protection effect

      目的  验证重组人骨形态发生蛋白10(rhBMP10)的心脏保护作用。
      方法  使用CHO细胞表达并纯化rhBMP10; 利用多柔比星多次给药构建小鼠心力衰竭模型; 使用超声心动图、TUNEL染色等方法评价小鼠心功能变化和心肌细胞凋亡情况。
      结果  本研究成功表达了具有生物活性的rhBMP10; 多柔比星造模小鼠的心功能受到严重损伤; 心肌细胞凋亡比例显著升高,而rhBMP10处理的小鼠心脏功能和心肌细胞未受到显著影响。
      结论  rhBMP10在减轻多柔比星引起的心脏损伤方面具有积极作用。

     

    Abstract:
      Objective  To verify the cardiac protection effect of recombinant human bone morphogenetic protein 10 (rhBMP10).
      Methods  The rhBMP10 was expressed and purified by CHO cells, heart failure model was established by repeated administration of doxorubicin, and cardiac function and apoptosis were evaluated by echocardiography and TUNEL staining.
      Results  In this study, rhBMP10 with biological activity was successfully expressed. Cardiac function of doxorubicin-induced mice was severely damaged. Apoptotic rate of cardiomyocytes significantly increased, and cardiac function and cardiomyocytes of rhBMP10-treated mice were not affected significantly.
      Conclusion  The rhBMP10 has a positive effect on reducing heart damage caused by doxorubicin.
