血清中微小RNA-506、微小RNA-934水平对胰腺癌患者术后生存结局的预测价值

Predictive value of serum microRNA-506 and microRNA-934 levels in postoperative survival outcome of patients with pancreatic cancer

    目的 探讨血清中微小RNA-506(miR-506)、微小RNA-934(miR-934)表达水平对胰腺癌患者术后生存结局的预测价值。
    方法 选取接受手术治疗的115例胰腺癌患者为研究组,并根据随访结果分为生存组(n=18)和死亡组(n=97)。选取同期就诊的50例胰腺良性病变患者为对照组; 选取同期体检健康的50例志愿者作为健康组。采用实时荧光定量聚合酶链反应(qRT-PCR)检测各组血清中miR-506和miR-934的相对表达水平。采用多因素Logistic回归分析法分析胰腺癌患者术后生存结局的影响因素。采用受试者工作特征(ROC)曲线分析血清miR-506、miR-934对胰腺癌患者术后生存结局的预测价值。
    结果 研究组患者的血清miR-506表达水平低于对照组,差异有统计学意义(P < 0.05)。研究组患者的血清miR-934表达水平高于对照组和健康组,差异有统计学意义(P < 0.05)。生存组的血清miR-506表达水平高于死亡组,血清miR-934表达水平低于死亡组,差异有统计学意义(P < 0.05)。miR-506、miR-934、TNM分期、淋巴结转移是胰腺癌患者术后是否生存的独立影响因素(P < 0.05)。
    结论 miR-506在胰腺癌术后死亡患者血清中低表达,miR-934在胰腺癌术后死亡患者血清中高表达。miR-506、miR-934预测胰腺癌患者术后生存结局的价值较高。

     

    Abstract:
    Objective To investigate the predictive value of serum microRNA-506 (miR-506) and microRNA-934 (miR-934) expression levels in postoperative survival outcomes of patients with pancreatic cancer.
    Methods A total of 115 patients with pancreatic cancer who received surgical treatment were selected as the study group, and were divided into survival group (n=18) and death group (n=97) according to the follow-up results. Fifty patients with benign pancreatic lesions in the same period were selected as control group; fifty healthy volunteers were selected as the healthy group. Quantitative real-time polymerase chain reaction (qRT-PCR) was used to detect the relative expression levels of miR-506 and miR-934 in serum of each group. Multivariate Logistic regression analysis was used to analyze the influencing factors of postoperative survival outcomes in patients with pancreatic cancer. The predictive value of serum miR-506 and miR-934 in postoperative survival outcome of pancreatic cancer patients was analyzed by receiver operating characteristic (ROC) curve.
    Results The expression level of serum miR-506 in the study group was significantly lower than that in the control group (P < 0.05). The expression level of serum miR-934 in the study group was significantly higher than that in the control group and healthy group (P < 0.05). The expression level of serum miR-506 in the survival group was significantly higher than that in the death group, and the expression level of serum miR-934 was significantly lower than that in the death group (P < 0.05). The miR-506, miR-934, TNM stage and lymph node metastasis were independent factors influencing postoperative survival of pancreatic cancer patients (P < 0.05).
    Conclusion The expression of miR-506 is low in the serum of patients who died after pancreatic cancer surgery, and the expression of miR-934 is high in the serum of patients who died after pancreatic cancer surgery. The value of miR-506 and miR-934 in predicting postoperative survival outcome of pancreatic cancer patients is high.
