Effects of endothelin receptor B on the contactile function of rat hepatic stellate cell

  • 摘要: 目的 观察内皮素受体B(ETB)激动后对培养鼠肝星状细胞收缩功能以及胞内钙(Ca2+)的影响.方法 分离培养大鼠HSC, 采用胶原晶格法观察ETA和ETB受体激动剂ET-1及选择性ETB受体协同剂IRI-1620对肝星状细胞收缩的影响.采用Fura-2/AM钙荧光指示剂测定HSC细胞内Ca2+浓度,并观察ETB对其影响.结果 选择性内皮素B受体协同剂IRL-1620在1 μmol/L浓度时即能明显抑制较低浓度的ET-1引起的肝星状细胞的收缩同时导致HSC细胞内Ca2+浓度的下降(P<0.05).结论 ETB被激动后可以抑制HSC的收缩,这种效应与ETB降低HSC细胞内Ca2+浓度有关.
