自杀基因疗法结合HSP-PC瘤苗抗黑色素瘤研究

ANTI-MELANOMA BY SUICIDE GENE THERAPY COMBINED WITH HSP-PC TUMOR VACCINE

  • 摘要: 目的 :研究 5 -氟尿嘧啶 /胞嘧啶脱氨酶 (5FC/CD)基因疗法与热休克蛋白 -多肽复合物 (HSP -PC)瘤苗免疫疗法联合抗肿瘤效果。方法 :将携带CD基因的重组腺病毒注射到小鼠黑色素瘤体内 ,腹腔注射 5 -FC ,同时皮下接种HSP70 -PC。结果 :经联合治疗后 ,70 %荷瘤小鼠肿瘤体积缩小、消退 ,小鼠存活期延长 ,肿瘤组织明显坏死 ,炎症细胞、CD4+ 、及CD8+ T细胞浸润明显。结论 :5 -FC/CD基因疗法结合HSP -PC瘤苗免疫疗法抗小鼠黑色素瘤作用显著 ,具有临床应用前景。

     

    Abstract: Objective:To investigate the antitumor effect of 5-fluorocytosine/cytosine deaminase (5FC/CD) suicide gene system combined with heat shock protein-peptide complexes(HSP-PC) tumor vaccine. Methods:Adenoviruses containing E.coli CD gene were injected into murine tumor followed by intraperitoneal murine 5-FC. Meanwhile, mice bearing tumor were inoculated s.c with HSP-PC.Results:In mice bearing tumor, the combined therapy led to reduction in tumor growth and complete tumor regression was observed in 70% mice. After the combined therapy, the tumor mass showed obvious necrosis and inflammatory cell infiltrcition, more CD 4 + and CD 8 +T cells infiltrated into the tumor tissue, and the murine CTL activities increased significantly. Conclusion:The combination therapy with 5-FC/CD system and HSP-PC tumor vaccine is very effective for murine melanoma, and has potential clinical useages.
