人脱钙骨基质粉诱导大鼠异位骨髓形成的初步研究

THE PRIMARY STUDY OF HUMAN DEMINERALIZED BONE MATRIX INDUCING ECTOPIC BONE MARROW FORMATION

  • 摘要: 目的 :建立诱导性骨髓形成的模型 ,探讨骨髓形成及调控机理。方法 :将异种 (人 )脱钙骨基质粉植入Spraque -Dawley大鼠腹壁皮下。结果 :在诱导 3~ 10d ,可见间充质细胞增殖 ,分化成幼稚软骨细胞并形成软骨组织。诱导11~ 2 1d即见到初级骨髓组织形成。诱导 2 2~ 3 4d出现典型骨髓组织 ,大量造血细胞产生。诱导 40~ 60d生成的造血组织未见明显萎缩。结论 :人鼠骨诱导活性因子具有同源性 ,无种的特异性 ,该模型建立可为临床血液学基础研究骨髓构形的发生学及其调控机理提供一个新的研究方向

     

    Abstract: Objective:To establish a model of inducing bone marrow formation with transplanting the heterogeneous demineralized bone matrix into Spraque-Dawley rat's bilateral abnominal subcutaneous tissue,to explore the mechanism of bone marrow formation and modulation.Methds:To transplant the heterogeneous(human) demineralized bone matrix into Spraque-Dawley rat's abdominal subcut-aneous tissue.Results:On 3rd to 10th day,mesenchymal cells proliferated,differentiated to immature cartilage cells and formed cartilage tissue.On 11th-21th day,primary bone marrow tissue formed.On 22th-34th day,typical bone marrow tissue appeared and a great quantity of hematopoietic cells were seen.On 4oth-60th day, no obvious atrophy of the induced hematopoitic tissue was seen.Conclusion:Bone inducing active factors of human and rat is homology.There is no herterogeneous.This model provides a new methord to study the mechanism of the formation and the modulation of bone marrow for clinical hematology.
