环状RNA_0032131靶向微小RNA-145-5p调控类风湿关节炎滑膜成纤维细胞的增殖、迁移和侵袭

Circular RNA_0032131 regulates the proliferation, migration and invasion of rheumatoid arthritis synovial fibroblasts by targeting microRNA-145-5p

    目的 探讨环状RNA_003213(hsa_circ_003213)靶向微小RNA-145-5p (miR-145-5p)对类风湿关节炎滑膜成纤维细胞(RA-FLSs)增殖、迁移和侵袭的影响。
    方法 检测RA患者和健康体检者外周血、RA-FLSs、人正常滑膜成纤维细胞中hsa_circ_0032131和miR-145-5p的水平。将RA-FLSs分为对照(NC)组、si-NC组、si-hsa_circ_0032131组、miR-NC组、miR-145-5p组、anti-miR-NC+si-hsa_circ_0032131组、anti-miR-145-5p+si-hsa_circ_0032131组、pcDNA组、pcDNA-hsa_circ_0032131组。采用CCK-8和Transwell法检测RA-FLSs活力、迁移和侵袭数。通过双荧光素酶报告实验确定hsa_circ_0032131与miR-145-5p的靶向关系。
    结果 与健康体检者相比, RA患者外周血中hsa_circ_0032131表达增加, miR-145-5p表达降低, 差异有统计学意义(P < 0.05)。与人正常滑膜成纤维细胞相比, RA-FLSs中hsa_circ_0032131表达增加, miR-145-5p表达降低,差异有统计学意义(P < 0.05)。与NC组比较, si-hsa_circ_0032131组RA-FLSs的活力和迁移、侵袭数目降低,差异有统计学意义(P < 0.05)。与miR-NC组比较, miR-145-5p组RA-FLSs的活力和迁移、侵袭数目降低,差异有统计学意义(P < 0.05)。与anti-miR-NC+si-hsa_circ_0032131组比较, anti-miR-145-5p +si-hsa_circ_0032131组RA-FLSs的活力和迁移、侵袭数目增高,差异有统计学意义(P < 0.05)。与pcDNA组比较, pcDNA-hsa_circ_0032131组RA-FLSs中miR-145-5p水平降低,差异有统计学意义(P < 0.05)。miR-145-5p是hsa_circ_0032131的直接靶点。
    结论 敲低hsa_circ_0032131通过上调miR-145-5p抑制RA-FLSs的增殖、迁移和侵袭。

     

    Abstract:
    Objective To investigate the effect of circular RNA_003213 (hsa_circ_003213) targeting microRNA-145-5p (miR-145-5p) on the proliferation, migration and invasion of rheumatoid arthritis synovialfibroblasts (RA-FLSs).
    Methods The levels of hsa_circ_0032131 and miR-145-5p in peripheral blood of RA patients and healthy subjects, RA-FLSs as well as normal human synovial fibroblasts were detected. RA-FLSs were divided into control (NC) group, si-NC group, si-hsa_circ_0032131 group, miR-NC group, miR-145-5p group, anti-miR-NC+si-hsa_circ_0032131 group, anti-miR-145-5p+si-hsa_circ_0032131 group, pcDNA group and pcDNA-hsa_circ_0032131 group. CCK-8 and transwell assays were used to detect the viability, migration and invasion of RA-FLSs. The targeting relationship between hsa_circ_0032131 and miR-145-5p was determined through a dual luciferase reporter assay.
    Results Compared with healthy subjects, the expression of hsa_circ_0032131 was increased, while the expression of miR-145-5p was decreased in the peripheral blood of RA patients, and the difference was statistically significant (P < 0.05). Compared with normal human synovial fibroblasts, the expression of hsa_circ_0032131 was increased, while the expression of miR-145-5p was decreased in RA-FLSs, and the difference was statistically significant (P < 0.05). Compared with the NC group, the viability, migration and the number of RA-FLSs invasion in the si-hsa_circ_0032131 group were reduced, and the difference was statistically significant (P < 0.05). Compared with the miR-NC group, the viability, migration and the number of RA-FLSs invasion in the miR-145-5p group were reduced, and the difference was statistically significant (P < 0.05). Compared with the anti-miR-NC+si-hsa_circ_0032131 group, the viability, migration and the number of RA-FLSs invasion in the anti-miR-145-5p+si-hsa_circ_0032131 group were increased, and the difference was statistically significant (P < 0.05). Compared with the pcDNA group, the level of miR-145-5p in the RA-FLSs of the pcDNA-hsa_circ_0032131 group was reduced (P < 0.05). The miR-145-5p was a direct target of hsa_circ_0032131.
    Conclusion Knockdown of hsa_circ_0032131 inhibits the proliferation, migration, and invasion of RA-FLSs by upregulating miR-145-5p.
