Abstract:
Objective To qualitatively analyze the water-soluble components in Bombyx Batryticatus by liquid chromatography tandem mass spectrometry (LC-MS/MS) technique and establish the analytical methods of fingerprint and common peaks identification.
Methods The column was Acclaim PA 25 μm C18 column (4.6 mm×250.0 mm), the mobile phase A and B were 0.2% phosphoric acid solution and acetonitrile respectively, the flow rate was 0.2 mL/min, the column temperature was 30℃, and the detection wave length was 265 nm. An electrospray ionization in both negative and positive ion modes was used to detect the molecular weight and fragment molecular of characteristic peaks obtained by LC-MS/MS, and characteristic peaks were identified.
Results A total of 19 common peaks were found in the fingerprints of water-soluble components of Bombyx Batryticatus, and 10 of them were identified by LC-MS/MS. The similarity evaluation results of fingerprints of 10 batches of Bombyx Batryticatus from different producing areas showed that the similarity was between 0.985 and 1.000.
Conclusion In this study, the method of fingerprint analysis based on LC-MS/MS is characterized by high selectivity, specificity and sensitivity, which can provide technical reference for quality evaluation of Bombyx Batryticatus.
