肺栓塞患者补体相关基因mRNA的表达特征

Expression characteristics of mRNA of complement-related genes in patients with pulmonary embolism

  • 摘要: 目的 分析症状性肺栓塞(PE)患者补体系统相关基因mRNA的表达特征。 方法 采用全人类基因组表达谱芯片,系统检测、分析20例PE患者与20例对照者补体系统相关基因mRNA的表达差异。 结果 (1) 补体固有成分基因共21条,与对照组相比, PE组C5 mRNA表达显著上调(P<0.05), C1QA、C1QB、C4B、CFP mRNA表达显著上调(P<0.01), C6 mRNA表达显著下调(P<0.01)。(2) 补体受体基因共7条, PE组中4条基因mRNA表达较对照组显著上调(P<0.01), 包括CR1、ITGAX、ITGAM、C5AR1。(3) 补体调节蛋白基因共7条,与对照组比较, PE组显著上调的基因有CD59(P<0.05)、CD55(P<0.01), CFI基因mRNA表达显著下调(P<0.05)。 结论 PE患者补体固有成分、受体及调节蛋白基因mRNA表达失衡,提示补体免疫功能紊乱和/或缺陷,最终导致补体介导的溶膜能力降低。

     

    Abstract: Objective To analyze the expression characteristics of mRNA of complement-related genes in patients with symptomatic pulmonary embolism(PE). Methods The whole human genome expression profile chip was used to analyze the expression differences of mRNA of complement-related genes in 20 PE patients and 20 controls. Results (1) There were 21 genes of complement intrinsic component. Compared with the control group, the expression of C5 mRNA increased significantly in PE group(P<0.05), the expressions of C1QA, C1QB, C4B and CFP mRNAs increased significantly in PE group(P<0.01), and the expression of C6 mRNA significantly decreased(P<0.01). (2) There were 7 complement receptor genes in PE group, and the mRNA expressions of CR1, ITGAX, ITGAM and C5AR1 in PE group were significantly higher than those in control group(P<0.01). (3) There were 7 complement regulatory protein genes in PE group. Compared with the control group, mRNA expressions of CD59(P<0.05)and CD55(P<0.01)increased significantly in PE group, while mRNA expression of CFI decreased significantly(P<0.05). Conclusion The imbalance of mRNA expression of complement intrinsic component, receptor and regulatory protein gene in PE patients suggests that the immune function of complement is disordered and(or)defective, which eventually leads to the decrease of membrane dissolving ability mediated by complement.
