长链非编码RNA KCNQ1重叠转录物1对肺鳞癌细胞增殖、迁移及侵袭能力的影响

Influence of long non-coding RNA KCNQ1 overlapping transcript 1 on cellular proliferation, migration and invasive ability of lung squamous cell carcinoma

      目的  检测长链非编码RNA(lncRNA) KCNQ1重叠转录物1(KCNQ1OT1)在肺鳞癌组织中的表达及其对肺鳞癌NCI-H266细胞增殖、迁移及侵袭能力的影响。
      方法  采用实时定量荧光PCR(RT-PCR)检测KCNQ1OT1基因在肺鳞癌组织中的表达。构建KCNQ1OT1基因的小干扰RNA(siRNA-KCNQ1OT1)并转染NCI-H266细胞, RT-PCR检测转染效果; MTT法检测NCI-H266细胞的增殖活力; 划痕实验检测NCI-H266细胞的迁移能力; Transwell小室检测NCI-H266细胞的侵袭能力。
      结果  KCNQ1OT1基因在肺鳞癌组织中呈显著高表达(P < 0.05), KCNQ1OT1的高表达与肺鳞癌分化程度、肿瘤体积、淋巴结转移以及TNM分期相关。si-KCNQ1OT1的转染能够显著下调NCI-H266细胞中KCNQ1OT1的表达(P < 0.05), 沉默KCNQ1OT1能够显著抑制NCI-H266细胞的增殖活力、迁移能力、侵袭能力(P < 0.05)。
      结论  KCNQ1OT1基因的表达上调与肺鳞癌的发生、发展相关,沉默KCNQ1OT1能够显著抑制NCI-H266细胞的增殖以及迁移和侵袭能力。

     

    Abstract:
      Objective  To detect the expression of long non-coding RNA (lncRNA) KCNQ1 overlapping transcript 1 (KCNQ1OT1) in tissues of lung squamous cell carcinoma and to explore the influence of KCNQ1OT1 gene on cellular proliferation, migration and invasive ability of NCI-H266 cells.
      Methods  Real-time PCR (RT-PCR) was used to detect the expression of KCNQ1OT1 gene in the tissues of lung squamous cell carcinoma. Small interfering RNA (siRNA-KCNQ1OT1) of KCNQ1OT1 gene was constructed to transfect the NCI-H266 cells, and RT-PCR was used to detect transfection effect. MTT was used to detect cellular proliferation of the NCI-H266 cells, scarification test was used to detect migration ability of the NCI-H266 cells, and the invasive ability of NCI-H266 cells was detected in Transwell cells.
      Results  KCNQ1OT1 gene was significantly over-expressed in lung squamous cell carcinoma (P < 0.05). The over-expression of KCNQ1OT1 was correlated with differentiation, tumor volume, lymph node metastasis and TNM stage of lung squamous cell carcinoma. The expression of KCNQ1OT1 in NCI-H266 cells was significantly down-regulated by transfection of si-KCNQ1OT1 (P < 0.05). Silencing KCNQ1OT1 was able to significantly inhibit the proliferation, migration and invasion of NCI-H266 cells (P < 0.05).
      Conclusion  Up-regulation of KCNQ1OT1 gene expression is associated with the occurrence and development of lung squamous cell carcinoma. Silencing KCNQ1OT1 can significantly inhibit the proliferation, migration and invasion of NCI-H266 cells.
