银屑病患者皮肤间充质干细胞中表皮生长因子、转化生长因子-β1的表达水平及其意义

Expression levels and significance of EGF,TGF-β1 of skin mesenchymal stem cells in patients with psoriasis

  • 摘要: 目的:观察银屑病患者皮肤间充质干细胞(SMSCs)的形态特征、免疫表型、诱导分化情况及其分泌表皮生长因子(EGF)及转化生长因子-β1(TGF-β1)的水平,尝试探讨银屑病可能的发病机制。方法选取35例银屑病及35例非银屑病患者设为观察组及对照组,分离、培养2组人群的 SMSCs,流式细胞仪鉴定免疫表型,多向分化法鉴定细胞分化功能,ELISA 法测定 EGF、TGF-β1的表达水平。结果2组患者的 SMSCs 形态学特征、免疫表型及诱导分化结果比较相似,观察组 SMSCs 分泌EGF 含量显著高于对照组(P <0.01),TGF-β1含量显著低于对照组(P <0.01),且2种细胞因子的含量高低与患者病情严重程度无相关性(P >0.05)。结论银屑病患者的 SMSCs 分泌 EGF、TGF-β1的水平异常,可能参与了银屑病的发病机制。

     

    Abstract: Objective To observe morphological characteristics,immunophenotyping,differ-entiation and the expression level of epidermal growth factor (EGF)and transforming growth factor-β1 (TGF-β1)in skin mesenchymal stem cells (SMSCs)so as to explore the possible mechanism of psori-asis.Methods 35 patients with psoriasis and 35 patients with non-psoriasis were selected and de-signed as observation group and control group.SMSCs of 2 groups were separated and cultured.Immu-nophenotyping was determined by flow cytometry (FCM).Cell differentiation ability was evaluated by multidirectional differentiation method.The expression level of EGF and TGF-β1 was detected by ELISA.Results Morphological characteristics,immunophenotyping,differentiation of SMSCs were similar.The expression level of EGF in observation group was higher than control group (P <0.01), but the expression level of TGF-β1 was significantly lower than control group(P <0.01).In addition, the expression level of EGF and TGF-β1 in 2 groups had no significant relationship with the severity of disease(P >0.05).Conclusion The expression level of EGF and TGF-β1 is abnormal in patients with psoriasis,which may participates in the pathogenesis of psoriasis.
